Cell culture laboratory should be able to carry out aseptic operation, culture, preparation, cleaning, disinfection and sterilization, storage and other six aspects of work.
1. Sterile operating area
(1) Aseptic work area: Aseptic work area is limited to aseptic work area such as cell culture. It is best to isolate from the outside world and not to cross or be disturbed by other factors.
The ideal sterile operating room should be divided into three parts:
1)The dressing room - changing clothes, shoes, hats and masks.
B) Buffer room - between the dressing room and the operating room. The purpose of this buffer room is to ensure the sterile environment of the operating room. At the same time, the thermostat and necessary small instruments can be placed.
C) Sterile operating room - specialized in aseptic operation, cell culture. Medium size, the top should not be too high (no more than 2.5m) to ensure the effective germicidal efficacy of ultraviolet radiation; smooth wall, no dead angle, easy to clean and disinfect. The tables should not be placed against the wall. The tables should be pressed flat on the plastic surface, coated with white or grey for anatomical tissue use, and observed the phenol red to show the pH value.
(2) Purification table: simple operation, convenient installation, small space, good purification effect. There are two kinds of purification platforms in bacterial culture room: a) side flow purification platforms or vertical purification platforms? B) external flow, or horizontal laminar flow.
2. Incubation area
The requirement for sterility in this area is not as strict as that in aseptic areas, but it still needs to be clean and dustless, so it should be set in areas with less interference, rather than in areas with heavy traffic. Incubation can be carried out in a thermostat or in a temperature-controlled greenhouse, which is expensive and is usually used in laboratories.
3. Preparatory area
This area is mainly engaged in the preparation of culture medium and related culture medium.
4. Storage area
Mainly store all kinds of refrigerators, drying boxes, liquid nitrogen tanks, sterile culture media, culture bottles, etc. This environment also needs to be clean and dust-free.
5. Clean, disinfect and disinfect areas
Cleaning and disinfection areas should be separated from other areas, mainly for cleaning, preparation, disinfection and preparation of three-steamed water for all cell culture containers.
